چکیده

25 MicroRNAs (miRNAs) are single stranded small RNA molecules that regulate various cellular 26 processes. miR-155 regulates various aspects of innate and adaptive immune response and plays 27 a key role in various viral infections and resulting neuroinflammation. In the present study, 28 evaluated the involvement of miR-155 in modulating Japanese Encephalitis Virus (JEV)29 induced neuroinflammation. We observed that miR-155 expression was up regulated during JEV 30 infection of mouse primary microglia, BV-2 microglia cell line, and in both mouse and human 31 brain. In vitro and in vivo knockdown of miR-155 minimized JEV-induced inflammatory 32 responses. In the present study, we confirmed targeting of SHIP1 3'UTR by miR-155 in the 33 context of JEV infection. Inhibition of SHIP1 by miR-155 resulted in higher interferon beta 34 (IFN) and pro-inflammatory cytokine production through activation of TANK-binding kinase 35 1 (TBK-1). Based on these observations we conclude that miR-155 modulates 36 neuroinflammatory response during JEV infection via negative regulation of SHIP1 expression. 37 Thus, modulation of miR-155 could be a novel strategy to regulate JEV induced 38 neuroinflammation. 39

Deficiency of Src homology 2 domain-containing inositol 5-phosphatase 1 affects platelet responses and thrombus growth.

Platelets are critical for normal hemostasis. Their deregulation can lead to bleeding or to arterial thrombosis, a primary cause of heart attack and ischemic stroke. Src homology 2 domain-containing inositol 5-phosphatase 1 (SHIP1) is a 5-phosphatase capable of dephosphorylating the phosphatidylinositol 3,4,5-trisphosphate second messenger into phosphatidylinositol 3,4-bisphosphate. SHIP1 plays...

Src homology 2 domain-containing inositol-5-phosphatase 1 (SHIP1) negatively regulates TLR4-mediated LPS response primarily through a phosphatase activity- and PI-3K-independent mechanism.

Src homology 2 (SH2) domain-containing inositol-5-phosphatase 1 (SHIP1) plays important roles in negatively regulating the activation of immune cells primarily via the phosphoinositide 3-kinase (PI-3K) pathway by catalyzing the PI-3K product PtdIns-3,4,5P3 (phosphatidylinositol-3,4,5-triphosphate) into PtdIns-3,4P2. However, the role of SHIP1 in Toll-like receptor 4 (TLR4)-mediated lipopolysacc...

Inositol phosphatase SHIP1 is a primary target of miR-155.

MicroRNA-155 (miR-155) has emerged as a critical regulator of immune cell development, function, and disease. However, the mechanistic basis for its impact on the hematopoietic system remains largely unresolved. Because miRNAs function by repressing specific mRNAs through direct 3'UTR interactions, we have searched for targets of miR-155 implicated in the regulation of hematopoiesis. In the pre...

The role of SHIP1 in T-lymphocyte life and death.

SHIP1 [SH2 (Src homology 2)-containing inositol phosphatase-1], an inositol 5-phosphatase expressed in haemopoietic cells, acts by hydrolysing the 5-phosphates from PtdIns(3,4,5)P(3) and Ins(1,3,4,5)P(4), thereby negatively regulating the PI3K (phosphoinositide 3-kinase) pathway. SHIP1 plays a major role in inhibiting proliferation of myeloid cells. As a result, SHIP1(-/-) mice have an increase...

SH2-containing inositol 5-phosphatases 1 and 2 in blood platelets: their interactions and roles in the control of phosphatidylinositol 3,4,5-trisphosphate levels.

Src homology domain 2-containing inositol 5-phosphatases 1 and 2 (SHIP1 and SHIP2) are capable of dephosphorylating the second messenger PtdIns(3,4,5) P3 (phosphatidylinositol 3,4,5-trisphosphate) and interacting with several signalling proteins. SHIP1 is essentially expressed in haematopoietic cells, whereas SHIP2, a closely related enzyme, is ubiquitous. In the present study, we show that SHI...
